Golgo 13 is a lightgun game based on the manga of the same name. It chronicles the exploits of a talented hitman named Duke Togo/Golgo 13 as he takes on a variety of tasks. A lightgun that resembles a semi-automatic sniper rifle is used to control the game. The player must kill single targets with limited ammunition and a hard time constraint, placing a far greater premium on accuracy than in other lightgun games of a similar nature. Sequences reminiscent of comic books are intercut with each mission, playing out a wider narrative.

The player's accuracy rating is 100% at the beginning of their career. The chosen assignment's goal is then shown, along with the target zone, time restriction, and ammunition supply. The player is rewarded with an on-screen performance display at the conclusion of each task. A substantial amount of money is added to the player's total for each task that is successfully finished, and they receive an additional bonus for hitting the target zone. However, the player's accuracy rating is deducted and they are then able to select the next task if they are unable to finish the objective or if the timer hits zero. When the player's accuracy rating drops to zero, the game is ended.

Five distinct missions are available at the beginning of the player's career, and they can be finished in any sequence. A fresh set of assignments is made accessible when the five have been completed. There are four rounds in all, with five assignments in each round getting progressively tougher. Only when all of the assignments have been finished may a special stage be chosen for the final round.
